In chemistry and materials science, an acid-triggered response occurs when protonation, hydrolysis, or other acid-dependent reactions cause a change in structure, solubility, or activity. These systems are designed to operate selectively in environments where the pH is lower than neutral, enabling targeted behavior.

Common mechanisms include hydrolysis of acid-labile bonds such as acetals and ketals, imines, and hydrazones; cleavage of tert-butyl esters; and protonation-driven changes in polymer charge that alter swelling, dissolution, or payload release. Many acid-triggered systems rely on pH as the trigger rather than a reactive chemical reagent, allowing activation in acidic environments such as specific cellular compartments or diseased tissues.

Applications span drug delivery, imaging, and smart coatings. In drug delivery, acid-triggered designs release therapeutics in acidic microenvironments found in tumor tissues or inside endosomes and lysosomes after cellular uptake. In imaging, acid-activated fluorescent probes can improve contrast in acidic compartments. Smart coatings and agricultural formulations also exploit acid-triggered dissolution or release to achieve controlled performance in response to pH changes.

Common design motifs include acid-labile linkers (for example, hydrazones and acetals), pH-responsive polymers, and protonatable amine or imidazole-containing units that alter solubility or membrane interactions under acidic conditions. Considerations in development include achieving selectivity for the target pH, ensuring stability at physiological pH, and tuning release or activation kinetics to match intended use.
