Ysc

Ysc refers to a gene cluster in pathogenic Yersinia species that encodes the Yersinia type III secretion system, commonly called the Ysc-Yop system. This secretion apparatus is a hallmark virulence factor carried on the bacterium’s virulence plasmid and is essential for translocating a repertoire of effector proteins, the Yops, into host cells. The Ysc-Yop system is a prototype of the injectisome type III secretion machines found in several Gram-negative pathogens.

Structure and mechanism

The Ysc-Yop system forms a syringe-like nanomachine that spans the bacterial inner and outer membranes. It includes a basal secretion apparatus powered by an ATPase (such as YscN), a needle (YscF), and a translocon complex that inserts into the host cell membrane (in association with translocator proteins such as YopB and YopD). The outer membrane component, including a secretin (such as YscC), completes the conduit for effector delivery. Secretion is tightly regulated by environmental cues, including temperature and calcium concentration, with translocation of Yop effectors into host cells modulated during contact with target cells.

Function and impact

Yops are a group of effector proteins that manipulate host signaling, cytoskeletal dynamics, and immune responses to promote bacterial survival and dissemination. The Ysc-Yop system enables Yersinia to establish infection in a variety of hosts and contributes to diseases ranging from enteric infections to bubonic and pneumonic plague in historical contexts. Beyond its role in pathogenesis, the Ysc-Yop system serves as a widely studied model for understanding bacterial Type III secretion systems and host–pathogen interactions.
