Ubikuitiniintijärjestelmän, often referred to as the ubiquitin-proteasome system (UPS), is a fundamental cellular process in eukaryotic organisms responsible for protein degradation. This intricate pathway plays a critical role in maintaining cellular homeostasis by removing damaged, misfolded, or no longer needed proteins. The system comprises two main components: ubiquitin, a small regulatory protein, and the proteasome, a large protein complex that acts as a cellular garbage disposal.

The process begins with the selective tagging of target proteins for degradation by the covalent attachment of ubiquitin molecules. This ubiquitination cascade involves a series of enzymes, including E1 (ubiquitin-activating enzyme), E2 (ubiquitin-conjugating enzyme), and E3 (ubiquitin ligase). E3 ligases are particularly important as they confer substrate specificity, recognizing and binding to specific proteins destined for destruction. Once a protein is polyubiquitinated, it is recognized by the 26S proteasome.

The 26S proteasome, a barrel-shaped structure, unfolds the ubiquitinated protein and threads it into its core, where it is cleaved into small peptides. These peptides are then further broken down into amino acids, which can be recycled by the cell for new protein synthesis. The ubiquitin molecules are also released and reused in subsequent degradation events. The UPS is involved in a wide array of cellular functions, including cell cycle regulation, DNA repair, immune response, and signal transduction. Dysregulation of the UPS is implicated in various diseases, such as cancer, neurodegenerative disorders, and autoimmune diseases.
