This enzyme plays a crucial role in the body's detoxification process, particularly in metabolizing bilirubin, a waste product formed from the breakdown of red blood cells.

Variations in the UGT1A1 gene can lead to reduced activity or complete absence of the UGT1A1 enzyme. This impaired function can result in a buildup of bilirubin in the blood, a condition known as hyperbilirubinemia. One of the most well-known conditions associated with UGT1A1 variations is Gilbert's syndrome. This is a common, mild, and usually harmless genetic condition characterized by intermittent mild jaundice, particularly during periods of stress, illness, or fasting. Another more serious condition is Crigler-Najjar syndrome, which is much rarer and can lead to severe, life-threatening jaundice if left untreated. UGT1A1 variations can also affect the metabolism of certain medications, such as irinotecan, potentially increasing the risk of adverse drug reactions.
