Thyminedimers

Thyminedimers are chemical compounds formed by the linkage of two thymine molecules, which are one of the four nucleotide bases in DNA. These dimers typically arise as a result of exposure to ultraviolet (UV) radiation, which induces covalent bonding between adjacent thymine bases in DNA strands. This formation of thymine dimers is a common form of DNA damage that can interfere with normal DNA replication and transcription processes.

In biological systems, thymine dimers are primarily of concern because they can cause mutations if left unrepaired. The most prevalent type of thymine dimer is the cyclobutane pyrimidine dimer (CPD), in which two thymine bases become covalently bonded through a cyclobutane ring. Another type, the (6-4) photoproduct, involves a bond between the 6th position of one thymine and the 4th position of the adjacent thymine. These lesions distort the DNA double helix, impeding the function of DNA polymerases and other enzymes.

Cells have evolved repair mechanisms to address thymine dimers, most notably nucleotide excision repair (NER). Enzymes recognize the abnormal structure, excise the damaged segment, and fill in the gap using the complementary strand as a template.

Understanding thymine dimers is significant in fields like photobiology, molecular biology, and cancer research, as UV-induced DNA damage is a major factor in skin cancers. In addition, thymine dimers serve as model systems for studying DNA repair mechanisms and the effects of UV radiation on genetic material.
