Tenyészetkészítést, also known as culture preparation or media preparation, is a fundamental process in microbiology and other biological sciences. It involves the creation of suitable environments, typically in the form of culture media, to support the growth and multiplication of microorganisms, cells, or tissues in a laboratory setting. The primary goal is to provide the necessary nutrients, moisture, and appropriate physical conditions for the target organisms to thrive.

The process usually begins with selecting or formulating a specific culture medium. These media can be liquid broths or solid agar-based preparations. The composition of the medium is crucial and depends on the specific requirements of the organism being cultured. It often contains a carbon source (like sugars), a nitrogen source (like amino acids or peptides), inorganic salts, vitamins, and growth factors. For solid media, a gelling agent like agar is added.

Ingredients are carefully weighed and dissolved in distilled or deionized water. The mixture is then sterilized, most commonly by autoclaving, which uses high pressure and temperature to kill any contaminating microorganisms. This sterilization step is critical to ensure that only the intended organism grows and that experimental results are not compromised by unwanted microbial contamination.

Once sterilized, the medium is often dispensed into appropriate containers such as petri dishes, test tubes, or flasks. For solid media, it is poured into sterile petri dishes and allowed to solidify. The prepared media can then be stored under appropriate conditions or used immediately for inoculation.
