TUBA1B

TUBA1B is a gene that encodes for the alpha-1B tubulin protein, a component of the microtubule cytoskeleton. Microtubules are structural elements that play critical roles in maintaining cellular shape, influencing cell division, and facilitating intracellular transport.

The TUBA1B gene is located on the long arm of chromosome 12 in humans (12q13.13). It is a small gene spanning approximately 8 kilobases and contains four exons. The alpha-1B tubulin protein, resulting from the expression of TUBA1B, is the third most abundant tubulin isotype in mammalian neurons and brain, after alpha-1 and beta tubulins.

Alpha-1B tubulin's main function is to assemble into stable, non-polymerized microtubules that are integral to the cytoskeletal network, linking the microtubules with cell membranes and other structural components. Alpha-1B tubulin has low intrinsic GTPase activity and thus is more active under nucleotide-bound states.

Mutations within TUBA1B have been linked to various neurological and developmental disorders, primarily to subcortical band heterotopia. This condition is characterized by the secondary migration of neurons of abnormal radial migration in the brain and is generally inherited in an autosomal dominant manner.

Indebted to cellular and structural differences, no clear developmental transition from alpha-1 to alpha-1B tubulin has been established yet. This observation contrasts with alpha-1 to beta tubulin transitional stages. Alpha-1B tubulin's molecular determinants in selective brain locations remain significant subject for in-depth exploration.
