It encompasses the translocase of the outer membrane (TOM) complex and the translocases of the inner membrane (TIM complexes), which together import the majority of nuclear-encoded mitochondrial proteins from the cytosol into the organelle.

The TOM complex sits in the outer membrane and includes a central pore-forming subunit and receptor components that recognize mitochondrial targeting signals. The TIM machinery resides in the inner membrane, with TIM23–TIM17 forming a presequence translocase for proteins bearing N-terminal targeting sequences, and TIM22 mediating the insertion of carrier proteins. Small TIM chaperones, such as Tim9 and Tim10, escort hydrophobic precursors through the intermembrane space, while matrix-side components like Tim44 and associated chaperones provide driving energy for import.

The import process begins with signal recognition at the mitochondrial surface, followed by translocation through TOM and transfer to the appropriate TIM complex. Protein import is powered by the electrochemical potential across the inner membrane (Δψ) and by ATP hydrolysis carried out by matrix chaperones such as mtHsp70, aided by Tim44. This coordinated pathway ensures correct protein targeting, folding, and maturation within the mitochondrial compartments.
