T2SS

T2SS, or Type II Secretion System, is a complex molecular machinery used by many Gram-negative bacteria to transport proteins from the bacterial interior across the cell envelope to the external environment. This secretion system plays a vital role in bacterial pathogenicity, environmental adaptation, and interaction with host organisms.

The T2SS is characterized by its multi-component structure, consisting of a membrane-embedded platform, a periplasmic pseudopilus, and an outer membrane secretin channel. It functions by recognizing specific substrate proteins, which are typically folded and often enzymatic, such as proteases, lipases, and toxins. The system harnesses energy, often derived from ATP hydrolysis, to assemble the pseudopilus, which acts like a piston to push substrates through the secretin pore outside the bacterial cell.

T2SS is evolutionarily related to the type IV pilus assembly system and shares similarities with the type IV secretion system (T4SS). It is widespread among pathogenic bacteria such as Pseudomonas aeruginosa, Vibrio cholerae, and Klebsiella spp., where it contributes to virulence by secreting factors that facilitate tissue invasion, immune evasion, and biofilm formation.

Research on T2SS has provided insights into bacterial secretion mechanisms and identified potential targets for antimicrobial development. Inhibiting T2SS function can impair bacterial ability to cause disease, making it a promising area for therapeutic intervention.
