SynthetaseRNase

SynthetaseRNase is a term used to describe a proposed or engineered bifunctional enzyme that combines aminoacyl-tRNA synthetase activity with ribonuclease activity in a single polypeptide or multi-domain complex. The concept leverages modular enzyme architecture to create a protein that can both activate an amino acid and process RNA substrates. While no natural enzyme of this exact class is established, researchers have explored the idea as a synthetic biology or protein engineering construct.

Typical designs place two catalytic regions in proximity: an aminoacyl-tRNA synthetase domain that catalyzes the ATP-dependent charging of a tRNA with a specific amino acid, and an RNase domain capable of endonucleolytic or exonucleolytic RNA cleavage. The two cores are linked by a flexible polypeptide or engineered docking interface, with an RNA-binding segment guiding substrate choice and orientation. Metal ions and ATP or other cofactors generally govern the respective activities.

Potential functions proposed for synthetaseRNases include coordination of RNA maturation with translation, quality control of tRNA pools, or targeted RNA turnover coupled to protein synthesis. In a research context, such hybrids could serve as tools for studying RNA processing or for designing multifunctional reagents in synthetic biology. Applications are speculative and depend on achieving compatible kinetics and regulatory control between activities.

Challenges include maintaining activity of both catalytic domains, preventing off-target RNA damage, avoiding interference between substrate channels, and ensuring proper cellular regulation. As of now, synthetaseRNase remains largely conceptual, with work focused on modular fusion constructs and in vitro demonstrations rather than established biological roles.
