Semiallogeneic

Semiallogeneic describes a genetic relationship between donor and recipient in transplantation and immunology in which the two individuals share some, but not all, histocompatibility antigens. The term is typically applied to tissues or cells that are not from the same individual (autologous) or fully genetically non-identical (allogeneic) but are partially compatible, most often in reference to major histocompatibility complex (MHC) antigens.

In practical terms, semiallogeneic grafts arise when donor and recipient are only partially histocompatible, such as individuals who share one haplotype (for example, certain parent-to-child or haploidentical donor–recipient pairs). Partial MHC matching can result in reduced, but not eliminated, immunologic recognition compared with fully mismatched grafts. Consequently, semiallogeneic grafts may exhibit longer survival and engraftment than completely mismatched allografts but remain subject to rejection and ongoing immune responses. Semiallogeneic models are frequently used in transplantation biology to study mechanisms of tolerance, chimerism, and the balance between graft rejection and graft-versus-host effects.

Clinical and research relevance varies by context. In hematopoietic stem cell transplantation, semiallogeneic or haploidentical donors are used when fully matched donors are unavailable, with ongoing strategies aimed at promoting tolerance and reducing graft-versus-host disease while preserving graft function. In laboratory studies, semiallogeneic systems help illuminate histocompatibility determinants and the biology of immune acceptance versus rejection.
