SLC34A1related refers to conditions, disorders, or phenotypes associated with mutations or alterations in the SLC34A1 gene. The SLC34A1 gene encodes the sodium-phosphate cotransporter 3 (NaPi-3), also known as NaPi-IIa, which is primarily expressed in the kidneys and intestines. This transporter plays a crucial role in the reabsorption of inorganic phosphate from the glomerular filtrate in the kidneys and absorption of phosphate from the diet in the intestines.

Mutations in the SLC34A1 gene can lead to a spectrum of clinical manifestations, collectively termed SLC34A1-related disorders. The most well-characterized condition associated with SLC34A1 mutations is hereditary hypophosphatemic rickets with hypercalciuria (HRH with hypercalciuria). This condition is characterized by impaired phosphate reabsorption in the kidneys, leading to hypophosphatemia (low phosphate levels in the blood). This, in turn, causes defective bone mineralization, resulting in rickets in children and osteomalacia in adults. The hypercalciuria (excess calcium in the urine) component can predispose individuals to kidney stone formation.

Other potential manifestations of SLC34A1-related conditions might include variations in phosphate homeostasis, renal function, and bone health, although these are less commonly described or may present with milder phenotypes. The inheritance pattern for SLC34A1-related disorders is typically autosomal recessive, meaning that two copies of the mutated gene are usually required for the condition to manifest. Diagnosis often involves biochemical testing of serum and urine phosphate and calcium levels, alongside genetic testing to confirm mutations in the SLC34A1 gene. Management focuses on addressing phosphate deficiency, bone deformities, and preventing kidney complications.
