Primersysteme, commonly referred to in English as primer systems, are collections of short nucleic acid sequences used to initiate synthesis of a target region in molecular biology. A primer system consists of oligonucleotides that are complementary to defined parts of a DNA or RNA template. By annealing to their targets, primers provide starting points for DNA synthesis in techniques such as polymerase chain reaction (PCR), sequencing, cloning, and diagnostic assays. Primer systems may be designed as a single pair for amplification or as multiplex panels containing multiple primer pairs.

Design and composition focus on specificity and efficiency. Typical primers are 18–30 nucleotides long, with balanced GC content and similar melting temperatures for primer pairs. They should avoid strong secondary structures and primer-dimer formation. In reverse transcription workflows, additional primers or adapters may be used. Sequencing applications often rely on primers that bind to known adapter or sequencing primer binding sites. Design tools (for example, Primer3, BLAST-based checks) help ensure target specificity and minimize off-target amplification.

Applications span gene expression analysis, pathogen detection, genotyping, and cloning. Primer systems enable targeted amplification, quantitative PCR, and initiation of sequencing reads. In multiplex designs, primer compatibility and assay dynamic range are critical considerations.

Limitations and validation include non-specific products, unequal primer efficiencies, and sensitivity to reaction conditions. Rigorous validation with controls, efficiency assessment, and, when applicable, standard curves are used to confirm performance before routine use. Primer systems are widely used in research and clinical diagnostics, and may be supplied as open sets or as vendor-specific panels.
