Pathwayfocused is a term used in bioinformatics and systems biology to describe an analytical and interpretive approach that centers biological pathways as the primary units of analysis. Unlike gene-centric analyses that focus on individual genes or proteins, pathwayfocused methods aim to assess the collective behavior and activity of predefined molecular pathways to understand phenotypes and disease mechanisms.

Conceptually, pathwayfocused relies on curated pathway collections such as KEGG, Reactome, and WikiPathways, and on computational methods that infer pathway activity from omics data. Common techniques include pathway enrichment analysis, which tests whether sets of genes linked to a pathway show coordinated changes; topology-aware approaches that incorporate the network structure within a pathway; and activity scoring or pathway-level condensation using methods like gene set variation analysis or network propagation.

Advantages include improved interpretability, reduced multiple testing burden, and the ability to capture coordinated responses that may be missed by single-gene analyses. Limitations involve dependence on the quality and coverage of pathway annotations, overlap between pathways leading to ambiguous results, bias toward well-studied pathways, and challenges in integrating heterogeneous data.
