Oncogeneactivated refers to a state where a gene that normally promotes cell growth and division has become abnormally active, contributing to cancer development. Oncogenes are altered versions of proto-oncogenes, which are essential for normal cellular processes. When proto-oncogenes undergo mutations or other genetic changes, they can transform into oncogenes. These oncogenes then promote uncontrolled cell proliferation, survival, and invasion, hallmarks of cancer.

The activation of oncogenes can occur through various mechanisms. These include point mutations that alter the protein's function, gene amplification leading to an overproduction of the protein, or chromosomal translocations that place the proto-oncogene under the control of a strong promoter or create a fusion gene with altered activity. For example, the BCR-ABL fusion gene, resulting from a translocation, is a well-known oncogene that drives chronic myeloid leukemia.

Once activated, oncogenes can disrupt the delicate balance of cell growth and death regulation. They can signal cells to divide continuously, resist programmed cell death (apoptosis), and promote the formation of new blood vessels (angiogenesis) to support tumor growth. The presence and activity of oncogenes are crucial targets for cancer diagnosis and therapy, with many anti-cancer drugs designed to inhibit the function of specific oncogenic proteins. Understanding oncogene activation is fundamental to comprehending the molecular basis of cancer.
