Nonsensemutaatiot, also known as nonsense mutations, are a type of gene mutation that results in the creation of a premature stop codon in the messenger RNA (mRNA) sequence. This occurs when a normal codon, which codes for an amino acid, is replaced by a stop codon (UAA, UAG, or UGA). Stop codons signal the termination of protein synthesis. Consequently, a nonsensemutaatio leads to the production of a truncated or shortened protein.

The effect of a nonsensemutaatio on an organism depends on various factors. If the premature stop codon occurs early in the gene, the resulting protein will be significantly shorter and may be completely non-functional. If the mutation occurs later in the gene, the truncated protein might retain some function, although it is often impaired. In some cases, the cell's quality control mechanisms can detect the aberrant mRNA and degrade it, preventing the synthesis of a potentially harmful protein. This process is known as nonsense-mediated decay.

They are a significant cause of genetic disorders. For example, cystic fibrosis and Duchenne muscular dystrophy are caused by nonsensemutaatiot in specific genes.
