Neuroligin4

Neuroligin4, often abbreviated as NLGN4 or NGL-4, is a protein encoded by the NLG4 gene. It is a member of the neuroligin family, which are transmembrane proteins crucial for synaptic function. Neuroligin4 is primarily expressed in the brain, with significant levels found in neurons.

The neuroligin family plays a key role in the formation and maturation of synapses, the specialized junctions between nerve cells where communication occurs. Neuroligin4, in particular, is thought to be involved in mediating the interaction between presynaptic and postsynaptic membranes. It binds to neurexins, another family of cell adhesion molecules located on the presynaptic side of the synapse, thereby helping to stabilize and organize synaptic structures.

Mutations in the NLG4 gene have been associated with certain neurodevelopmental disorders, most notably autism spectrum disorder (ASD) and X-linked intellectual disability. These genetic alterations can disrupt the normal function of NLGN4, leading to impaired synaptic signaling and connectivity, which are believed to contribute to the cognitive and behavioral characteristics of these conditions. Research into the precise mechanisms by which NLGN4 dysfunction contributes to these disorders is ongoing.
