In biology, cell lysis describes the bursting of a cell due to osmotic pressure, mechanical disruption, or enzymatic/chemical agents. Hemolysis is a well-known form of lysis affecting red blood cells, releasing hemoglobin into surrounding fluid. Lysis can occur naturally as part of immune responses, during viral infections, or as a controlled step in laboratory protocols. The complement system can also trigger lysis of targeted cells through membrane attack complexes. Osmotic lysis occurs when cells are placed in hypotonic environments, while chemical lysis uses detergents or enzymes to solubilize membranes.

In virology and bacteriology, lysis is central to the lytic cycle of many bacteriophages, wherein a phage replicates within a bacterium and then lyses the host to release progeny virions. This process usually involves holin proteins creating pores in the membrane and endolysins degrading the cell wall. Lytic cycles are contrasted with lysogeny, where the viral genome persists without immediate destruction of the host.

In the laboratory, lysis is employed for sample preparation, DNA and protein extraction, and cell disruption. Common approaches include enzymatic lysis (eg, lysozyme), chemical lysis with detergents (eg, SDS, Triton X-100), and physical methods (sonication, homogenization). Tools such as LDH-release assays monitor cellular lysis, while plaque assays quantify lytic activity of phages. Lysis-related techniques require careful control to preserve biomolecules and avoid undesired degradation.
