Kromatinmódosító, also known as chromatin modifiers, are a class of enzymes that play a crucial role in regulating gene expression by altering the structure of chromatin. Chromatin is the complex of DNA and proteins that forms chromosomes within the nucleus of eukaryotic cells. Its structure is not static but undergoes dynamic changes, and chromatin modifiers are key players in orchestrating these changes.

These enzymes achieve their function through various mechanisms. Some, like histone acetyltransferases (HATs), add acetyl groups to histone proteins, which are the primary protein components of chromatin. This acetylation generally loosens chromatin structure, making DNA more accessible to transcription machinery and thus promoting gene activation. Conversely, histone deacetylases (HDACs) remove these acetyl groups, leading to a more condensed chromatin structure and repression of gene transcription.

Other chromatin modifiers target different aspects of chromatin. For instance, histone methyltransferases (HMTs) add methyl groups to histones, and the effect of this modification can be either activation or repression of gene expression depending on the specific amino acid residue being methylated and the degree of methylation. ATP-dependent chromatin remodelers are another important group that use the energy from ATP hydrolysis to physically reposition, eject, or restructure nucleosomes, the basic units of chromatin.

The proper functioning of chromatin modifiers is essential for normal cellular processes, including development, differentiation, and response to environmental cues. Dysregulation of these enzymes has been linked to a variety of diseases, particularly cancer, where aberrant gene expression can drive uncontrolled cell growth and proliferation. Consequently, chromatin modifiers are an active area of research, with efforts focused on understanding their precise roles and developing targeted therapies.
