Kiinteäfaasitekniikat, commonly known in English as solid-phase techniques, are a broad category of chemical and biochemical methods where a reaction or purification process occurs on a solid support. This solid support, often a polymer resin or silica-based material, is functionalized with specific chemical groups or immobilized biomolecules. The key advantage of these techniques lies in the ease of separation of the solid phase from the liquid phase. This separation allows for efficient washing away of excess reagents and byproducts, simplifying purification steps and often leading to higher yields and purity of the desired product.

One of the most prominent applications of kiinteäfaasitekniikat is in solid-phase peptide synthesis (SPPS). In SPPS, amino acids are sequentially added to a growing peptide chain anchored to a resin. After each coupling step, the resin is washed, removing unreacted amino acids and coupling reagents. This process is highly automated and has revolutionized peptide production. Similar principles are applied in solid-phase organic synthesis (SPOS) for the construction of various organic molecules.

Another important area is solid-phase extraction (SPE), a powerful sample preparation technique. In SPE, analytes from a liquid sample are retained on a solid sorbent material, while interfering substances are washed away. The retained analytes are then eluted with a different solvent, achieving separation and concentration. Kiinteäfaasitekniikat are also widely used in affinity chromatography, where a ligand is immobilized on a solid support to selectively bind and purify a target molecule, such as a protein or antibody. The versatility and efficiency of these techniques have made them indispensable tools in research, diagnostics, and industrial processes.
