Inaktiveringsmetod refers to any process that aims to render a biological agent, toxin, or active substance non-viable or non-functional. The purpose is safety and control, enabling handling, study, or use of materials without risk of infection or unwanted activity. Inaktivering can target whole organisms, viral particles, toxins, enzymes, or genetic material. The approach is chosen based on the desired end point, stability of the substance, and risk considerations.

Common physical methods include heat inactivation (such as pasteurization, boiling, or autoclaving) and irradiation (UV or gamma rays). Chemical methods include aldehydes and alkylating agents (formaldehyde, glutaraldehyde), beta-propiolactone, alcohols, oxidizing agents, and detergents. Changes in pH can denature proteins or degrade nucleic acids; enzymatic inactivation uses proteases or nucleases. Inactivation can also be achieved by removal of activity through filtration or sequestration, though this is often considered decontamination rather than inactivation.

Applications include the development of inactivated vaccines, detoxification of toxins to form toxoids for vaccines, preparation of safe reference materials in diagnostic laboratories, and environmental or equipment decontamination. In serology, serum may be heat-inactivated to deactivate complement before certain assays.

Key considerations are preserving essential characteristics (for example, antigenicity in vaccines) while ensuring complete loss of pathogenicity or activity; avoiding formation of harmful by-products; addressing residual toxicity; and ensuring process validation and regulatory compliance.
