Immunohistochemistry (IHC) is a laboratory technique used to detect the presence of specific antigens in tissue sections. This method combines immunology and histology, allowing for the visualization of proteins within cells and tissues. The process involves several key steps: tissue fixation, sectioning, antigen retrieval, blocking, primary antibody incubation, secondary antibody incubation, and visualization. The primary antibody is specific to the antigen of interest, while the secondary antibody is conjugated with an enzyme or fluorescent dye that produces a visible signal.

IHC is widely used in medical research and diagnostics to study the distribution and expression of proteins in diseased tissues. It provides valuable information about the localization, quantity, and activity of proteins, which can aid in the diagnosis of diseases, the evaluation of treatment responses, and the understanding of disease mechanisms. The technique can be performed using various detection systems, including peroxidase, alkaline phosphatase, and fluorescent dyes, each offering different advantages in terms of sensitivity, specificity, and ease of use.

One of the main advantages of IHC is its ability to provide high-resolution images of protein expression within tissue sections. This allows researchers and clinicians to examine the spatial distribution of proteins at the cellular and subcellular levels. Additionally, IHC can be combined with other techniques, such as in situ hybridization and fluorescence in situ hybridization, to study the expression of multiple targets simultaneously.

However, IHC also has limitations. The technique relies on the availability of specific antibodies, which may not be readily available for all antigens. Additionally, the interpretation of IHC results can be subjective and may be influenced by factors such as tissue quality, antibody specificity, and detection system sensitivity. Despite these challenges, IHC remains a valuable tool in biomedical research and clinical practice, providing insights into the molecular and cellular basis of diseases.
