IDHStatus

IDHStatus is a term used in biomedical data annotation to denote the mutation status of isocitrate dehydrogenase genes, primarily IDH1 and IDH2, in a biological sample. The status is usually categorized as Mutant or Wild-Type, with possible sublabels such as IDH1_mutant or IDH2_mutant when a specific gene is mutated. In some datasets, samples may be annotated as IDH_NonMutant or IDH_Mutant to reflect composite results where both genes are evaluated.

IDHStatus can be established by genetic sequencing of IDH1 and IDH2, by targeted assays, or by immunohistochemistry for the IDH1 R132H mutation. Not all IDH mutations are detectable by IHC; sequencing provides broader detection.

IDH mutation status is a major molecular marker in certain cancers, especially lower-grade gliomas and some intrahepatic cholangiocarcinomas. IDH mutations are associated with distinct metabolic profiles, including accumulation of 2-hydroxyglutarate, and generally correlate with better prognosis and different therapeutic responses compared to IDH-wild-type tumors. In clinical guidelines, IDHStatus informs histopathological classification and may influence treatment planning.

In research databases and cancer registries, IDHStatus is used as a categorical variable to stratify analyses, compare outcomes, and harmonize cross-study datasets. Accuracy depends on assay sensitivity and reference databases; discrepancies can arise from partial testing or variants of uncertain significance.
