These tests are crucial for diagnosing and monitoring a range of conditions, including hemophilia, von Willebrand disease, and other inherited or acquired bleeding disorders. They are also used to assess the risk of excessive bleeding before surgery or in patients taking anticoagulant medications.

The process typically involves drawing a blood sample, which is then mixed with an anticoagulant to prevent clotting in the tube. The plasma is separated from the blood cells through centrifugation. In the laboratory, the plasma is incubated with reagents that activate specific clotting pathways, and the time it takes for a clot to form is measured. This clotting time is then compared to a reference range to determine the activity level of the particular coagulation factor being tested. Different assays exist to measure individual factors, such as Factor VIII, Factor IX, and von Willebrand factor. The results help clinicians identify which specific factor is deficient or dysfunctional, guiding appropriate treatment decisions.
