Hypothalamohypofysestien, also known as the hypothalamo-pituitary axis or the HPA axis, is a complex neuroendocrine system that controls the body's response to stress and regulates many other essential bodily functions. It involves the hypothalamus, a small region in the brain, and the pituitary gland, a pea-sized gland located at the base of the brain. These two structures work together in a feedback loop to maintain homeostasis, the body's internal balance.

The hypothalamus produces releasing and inhibiting hormones that act on the anterior pituitary gland. In response, the anterior pituitary secretes tropic hormones that stimulate other endocrine glands, such as the adrenal glands, thyroid gland, and gonads. The posterior pituitary, on the other hand, releases hormones like antidiuretic hormone (ADH) and oxytocin, which are produced in the hypothalamus and stored in the posterior pituitary.

The HPA axis plays a crucial role in the stress response. When the body perceives a threat, the hypothalamus releases corticotropin-releasing hormone (CRH), which signals the pituitary to release adrenocorticotropic hormone (ACTH). ACTH then prompts the adrenal glands to release cortisol, a stress hormone. Cortisol mobilizes energy resources and prepares the body for action. Once the stressor is removed, the increased levels of cortisol provide negative feedback to the hypothalamus and pituitary, shutting down the HPA axis. Dysfunction of the HPA axis can lead to various health problems, including Cushing's disease and Addison's disease, as well as mood disorders like depression and anxiety.
