Hemaglutinointitestit, often referred to as HA tests, are laboratory assays used to detect the presence of specific antibodies or antigens in a sample. The underlying principle relies on the ability of certain substances, most commonly viruses, to agglutinate or clump red blood cells. Agglutination is a visible process where red blood cells, in the presence of the agglutinating agent, form a lattice-like structure.

In a typical hemagglutination assay, a sample suspected of containing an antigen is mixed with a suspension of red blood cells. If the antigen is present and has hemagglutinating properties, it will bind to receptors on the surface of the red blood cells, causing them to clump together. This clumping is observed as a uniform suspension of red blood cells. Conversely, if the antigen is absent, the red blood cells will settle to the bottom of the test well or tube, forming a distinct pellet.

Hemagglutination inhibition (HI) tests are a variation of this assay and are widely used to detect specific antibodies. In an HI test, a known amount of a hemagglutinating agent, such as a virus, is pre-incubated with a sample suspected of containing antibodies. If specific antibodies are present, they will bind to the hemagglutinating agent, neutralizing its ability to cause agglutination. This mixture is then added to red blood cells. If agglutination is inhibited (i.e., no clumping occurs and the red blood cells settle), it indicates the presence of specific antibodies in the sample. HI tests are crucial in serological studies for diagnosing viral infections, monitoring immune responses to vaccines, and identifying viral strains.
