Grb2SOS

Grb2SOS is a protein complex involved in cellular signaling pathways that regulate cell growth, differentiation, and survival. The name derives from its two primary components: Growth factor receptor-bound protein 2 (Grb2) and Son of Sevenless (SOS). This complex plays a critical role in transmitting signals from receptor tyrosine kinases (RTKs) to the Ras GTPase, a key molecule in the mitogen-activated protein kinase (MAPK) cascade.

Grb2 is an adaptor protein that lacks intrinsic enzymatic activity but facilitates interactions between other signaling molecules. It contains an N-terminal SH3 domain, a central SH2 domain, and a C-terminal SH3 domain, which enable it to bind to phosphorylated tyrosine residues on activated RTKs and other proteins. SOS, on the other hand, is a guanine nucleotide exchange factor (GEF) that activates Ras by promoting the exchange of GDP for GTP on Ras proteins. When Grb2 binds to a phosphorylated RTK, it recruits SOS to the plasma membrane, where Ras resides, enabling SOS to stimulate Ras activation.

The Grb2-SOS complex is essential for various physiological processes, including embryonic development, cell proliferation, and neuronal growth. Dysregulation of this pathway has been implicated in several diseases, particularly cancers, where overactivation of Ras and downstream MAPK signaling can lead to uncontrolled cell growth. Mutations or amplifications in components of this pathway, such as RTKs or Ras itself, are commonly observed in tumors, highlighting its importance in oncogenesis.

Research on Grb2-SOS has also contributed to the development of targeted therapies aimed at disrupting aberrant signaling in cancer cells. Understanding the molecular interactions within this complex has provided insights into potential drug targets for treating diseases driven by dysregulated growth signaling.
