Gamma-tubulin-based refers to cellular processes and structures that rely on gamma-tubulin for the nucleation and organization of microtubules. Gamma-tubulin is a member of the tubulin superfamily that concentrates at microtubule organizing centers to initiate microtubule assembly, particularly at minus ends.

The main engine of gamma-tubulin-based nucleation is the gamma-tubulin ring complex (gamma-TuRC), which templates the formation of microtubules. Gamma-TuRC is composed of gamma-tubulin and multiple gamma-tubulin complex proteins (GCP2–GCP6), along with accessory factors such as NEDD1 and other scaffold proteins that help recruit the complex to nucleation sites. A smaller, core subcomplex called gamma-tubulin small complex (gamma-TuSC), made of gamma-tubulin with GCP2 and GCP3, can assemble into the larger gamma-TuRC in many organisms to enhance nucleation efficiency.

Localization of gamma-tubulin-based nucleation is tied to microtubule organizing centers (MTOCs). In animal cells, centrosomes serve as primary MTOCs where gamma-TuRC concentrates to nucleate microtubules during cell division. In plants and some fungi, nucleation can occur at alternative sites such as the nuclear envelope or spindle poles, reflecting variation in MTOC organization across lineages. Regulation of gamma-tubulin-based nucleation involves cell cycle cues and regulatory proteins (for example CDK-dependent factors) that control the timing and placement of gamma-TuRC at MTOCs.

Gamma-tubulin-based nucleation is essential for spindle formation, chromosome segregation, and proper cell division. Disruption of gamma-tubulin function or mislocalization can lead to defects in microtubule networks and aneuploidy, with implications for development and disease. The mechanism is highly conserved across eukaryotes and remains a focus of research on cytoskeletal organization.
