Eritadenine

Eritadenine is a natural product classified as a nucleoside-like compound that has been isolated from basidiomycete mushrooms, most notably the shiitake mushroom (Lentinula edodes). It has attracted scientific interest because of its potential effects on lipid metabolism.

Mechanism of action studies indicate that eritadenine inhibits S-adenosylhomocysteine hydrolase (SAH hydrolase). Inhibition of this enzyme can elevate intracellular S-adenosylhomocysteine levels, which in turn impairs S-adenosylmethionine-dependent methyltransferases. The resulting disruption of hepatic methylation reactions is associated with changes in lipid synthesis and secretion, contributing to observed reductions in serum cholesterol in some experimental models.

Biological activity and research context: Most evidence for the cholesterol-lowering effects of eritadenine comes from animal studies and in vitro experiments. In rodents, administration of eritadenine or mushroom extracts containing the compound has been associated with lowered total cholesterol and low-density lipoprotein cholesterol, with varying effects on triglycerides. Human data are limited, and there is no regulatory approval for eritadenine as a treatment or approved nutraceutical for cholesterol management.
