Dickkopfrelated

Dickkopfrelated refers to proteins that belong to the Dickkopf (DKK) family or are functionally associated with their pathways. The DKK family comprises secreted proteins that play crucial roles in embryonic development and adult tissue homeostasis, primarily by inhibiting the Wnt signaling pathway. There are four known members in mammals: DKK1, DKK2, DKK3, and DKK4. These proteins act as antagonists of Wnt signaling by binding to the Wnt receptor complex, specifically the low-density lipoprotein receptor-related proteins (LRP5 and LRP6), thereby preventing downstream signaling events.

The Dickkopfrelated proteins are involved in a wide range of biological processes. In development, they are critical for patterning of various tissues, including the central nervous system, limbs, and vertebrae. Consequently, DKK1 has become a target for therapeutic interventions in conditions like osteoporosis and bone metastases.

Beyond bone, Dickkopfrelated proteins are also implicated in the pathogenesis of various cancers. Their role in cancer is complex and can be context-dependent, with some DKK proteins acting as tumor suppressors and others as promoters. They can influence tumor cell proliferation, survival, invasion, and angiogenesis by modulating Wnt signaling in both cancer cells and the tumor microenvironment.
