Densitometern

A densitometer is an optical instrument used to quantify the optical density of a material by measuring how much light is transmitted through or reflected from it. It is employed to determine how strongly a sample absorbs or blocks light, providing a numerical value that helps compare different samples or monitor processes.

In transmission densitometry, light from a source passes through the sample and the detector measures the transmitted light. In reflection densitometry, light is directed at the surface and the detector measures the light reflected back. Results are usually expressed as optical density (OD) or percent transmission, with optical density following a logarithmic relationship: OD = log10(I0/I). Some densitometers perform color densitometry, measuring density across multiple color channels (for example red, green, and blue) to assess color balance.

Applications of densitometry are diverse. In printing and photography, densitometers monitor film densities and printed output to ensure color accuracy and repeatable quality control. In biochemistry and molecular biology, densitometry analyzes bands on gels or blot membranes after staining or chemiluminescent detection. In medicine, the term is also used for devices that measure bone mineral density, as in dual-energy X-ray absorptiometry (DEXA), though these are specialized densitometers for clinical assessment. Specialized instruments such as spectrodensitometers provide high-precision color measurements for packaging and calibration.

Historically, densitometers played a key role in color management and quality control, with brands and models evolving from early optical devices to modern digital and scanning densitometers. Limitations include calibration drift, sample nonuniformity, stray light, and a limited dynamic range, though digital imaging and software have expanded accessible densitometry beyond dedicated hardware.
