Chromatography-olfactometry, often abbreviated GC-O, is an analytical technique that combines chromatographic separation with human olfactory detection to identify odor-active compounds in complex mixtures. The method pairs a gas chromatograph with an olfactory detection port, enabling sensory evaluation of eluting compounds while instrumental data are collected.

In GC-O, the effluent from the GC splits between a conventional detector (such as a flame ionization detector or mass spectrometer) and a sniffing port where trained panelists inhale the aroma as peaks emerge. Panelists describe odor quality, note when an odor is detectable, and rate intensity on a defined scale. Retention indices and mass spectra are used to correlate sensory events with chemical identity, and aroma-focused techniques such as aroma extract dilution analysis (AEDA) may be employed to estimate odor potency.

The sniffing port may be coupled to an olfactometer that standardizes the odor stimulus and minimizes carryover and adaptation. Fresh, dehumidified carrier gas and careful panel training help improve reproducibility. Data are typically integrated with GC-MS or GC-FID results to assign odor descriptors to specific compounds or to odor-active regions of the chromatogram.

Applications of GC-O span flavor and fragrance research, essential oil characterization, food and beverage aroma profiling, environmental odor assessment, and pharmaceutical or agricultural analyses. Limitations include subjectivity and panel variability, odor fatigue, and the need for standardized protocols and experienced judges. Despite these challenges, chromatography-olfactometry remains a key method for linking chemical identity with perceived odor.
