Ceramide-related refers to topics and substances linked to ceramides, a family of bioactive lipids that are key components of sphingolipids in animal cells. Ceramides consist of a fatty acid attached to a sphingosine base and are especially abundant in the skin’s stratum corneum, where they help form a moisture-retaining barrier. They also occur throughout cell membranes and participate in various signaling pathways.

Biosynthesis and metabolism of ceramides occur via several pathways. The de novo pathway begins with serine palmitoyltransferase and proceeds through several intermediates to form ceramide. Ceramide synthases (CerS1–CerS6) introduce specific fatty acid chains, yielding a diversity of ceramides by chain length. Ceramides can also be generated by hydrolysis of sphingomyelin through sphingomyelinases or through salvage pathways from sphingosine. Ceramides are further metabolized by ceramidases into sphingosine, which can be phosphorylated to sphingosine-1-phosphate, a related lipid with opposing signaling effects. The relative activities of these enzymes influence cellular ceramide levels and, consequently, cell fate decisions.

Functions and signaling roles of ceramides are diverse. They contribute to membrane structure and the formation of ceramide-enriched microdomains that regulate receptor signaling. As bioactive mediators, ceramides can promote cell cycle arrest, differentiation, or apoptosis in response to stress. The balance between ceramide and sphingosine-1-phosphate signaling is often described as a rheostat that influences cellular outcomes.

Clinical and applied relevance includes skin and cosmetic science, where ceramides help restore the skin barrier and reduce transepidermal water loss. In medicine and research, ceramide metabolism is studied in relation to metabolic, neurodegenerative, and inflammatory diseases, and ceramide-based therapies and formulations are an active area of investigation.
