ATPase-based refers to processes powered by ATP hydrolysis carried out by ATPases. These enzymes convert the chemical energy of ATP into mechanical work, ion transport, or conformational changes essential for cellular function. The term covers ion pumps, molecular motors, and energy-conversion machines that rely on ATPase activity to operate.

Major ATPase families include P-type ATPases (ion pumps that form a phosphorylated intermediate), such as Na+/K+-ATPase and Ca2+-ATPase, which maintain ion gradients; and F-type and V-type ATPases, rotary enzymes that synthesize ATP using a proton motive force (F-type) or hydrolyze ATP to move protons (V-type). ABC transporters use ATP binding and hydrolysis to move diverse substrates across membranes.

Molecular motors such as myosin, dynein, and kinesin are ATPases whose hydrolysis of ATP powers movement along cytoskeletal tracks. ATPase-based mechanisms underpin muscle contraction, vesicle transport, and neuronal signaling, as well as energy metabolism in mitochondria and chloroplasts.

Applications and study areas include pharmacology (targeting ion pumps and transporters), biotechnology, and the design of synthetic molecular machines. Regulation occurs at transcriptional and post-translational levels, and malfunction of ATPases is associated with hypertension, cardiac disease, and metabolic disorders.
