AKTmediated

AKTmediated refers to biological processes or signaling pathways that are regulated or influenced by the protein kinase AKT, also known as Protein Kinase B (PKB). AKT is a serine/threonine kinase that plays a crucial role in various cellular functions, including cell survival, proliferation, metabolism, and growth. It is a key component of the phosphoinositide 3-kinase (PI3K)/AKT signaling pathway, which is frequently dysregulated in cancer and other diseases.

AKTmediated signaling is typically initiated by growth factors or other extracellular stimuli that bind to receptor tyrosine kinases or G protein-coupled receptors on the cell surface. This activation cascade often leads to the recruitment and activation of PI3K, which phosphorylates phosphatidylinositol (4,5)-bisphosphate (PIP2) to generate phosphatidylinositol (3,4,5)-trisphosphate (PIP3). PIP3 then recruits and activates AKT at the plasma membrane. Once activated, AKT can phosphorylate a wide range of downstream substrates, including proteins involved in apoptosis (e.g., BAD), cell cycle progression (e.g., p21, p27), and glucose metabolism (e.g., GSK3β, GLUT4).

Dysregulation of AKTmediated pathways is implicated in numerous pathological conditions. In cancer, hyperactivity of the PI3K/AKT pathway can promote tumor growth, survival, and resistance to therapy. Consequently, targeting AKTmediated signaling is an active area of research for developing novel cancer therapeutics. Beyond cancer, AKT is also involved in conditions like diabetes, neurodegenerative diseases, and cardiovascular disorders, highlighting its central role in cellular homeostasis.
