ReticulumtoGolgi refers to the process by which proteins and lipids synthesized in the endoplasmic reticulum (ER) are transported to the Golgi apparatus for further modification and sorting. This vesicular transport system is crucial for the proper functioning of the cell, ensuring that these molecules reach their designated destinations within or outside the cell.

The journey begins with the budding of transport vesicles from the ER. These vesicles encapsulate newly synthesized proteins and lipids. Proteins destined for secretion, membrane insertion, or delivery to other organelles are often folded and modified within the ER lumen. Lipids are also synthesized and assembled in the ER membrane. Once packaged into vesicles, these cargo molecules are then transported to the Golgi apparatus.

This transport occurs via a network of tubules and vesicles that bridge the ER and Golgi. Specific protein complexes, such as coat proteins like COPII, are involved in the formation and budding of these vesicles from the ER exit sites. These vesicles then move along cytoskeletal tracks towards the Golgi.

Upon reaching the Golgi, the vesicles fuse with the cis-Golgi network, the entry face of the Golgi apparatus. Here, the cargo is released and begins its passage through the different cisternae of the Golgi – the cis, medial, and trans Golgi. Within the Golgi, proteins undergo further processing, including glycosylation, phosphorylation, and proteolytic cleavage, while lipids are also modified. Finally, in the trans-Golgi network, the modified molecules are sorted and packaged into new vesicles for transport to their final destinations, such as lysosomes, the plasma membrane, or for secretion out of the cell. The reticulumtoGolgi pathway is thus a fundamental mechanism in cellular protein and lipid trafficking.
