Reporter assays are molecular biology techniques that use a reporter gene as a measurable readout of regulatory element activity. A reporter gene encodes an easily detected product, such as an enzyme or fluorescent protein, and is placed under the control of a promoter, enhancer, or other regulatory sequence. The amount or activity of the reporter product reflects the transcriptional activity of the regulatory element, allowing researchers to quantify promoter strength, enhancer activity, transcription factor function, or signaling pathway responses.

Common reporters fall into several categories. Enzymatic reporters include luciferases (firefly and Renilla), beta-galactosidase, and beta-glucuronidase, typically measured by luminescence or colorimetric assays. Fluorescent reporters such as GFP and its derivatives enable direct visualization and fluorometric measurements. Secreted reporters like SEAP can be measured in culture medium. Many experiments use dual-reporter systems, combining a primary reporter with an internal control reporter to normalize for transfection efficiency and cell number.

Experimental design typically involves constructing a plasmid where the regulatory element drives the reporter gene, followed by transfection into cells or introduction into organisms. Researchers often include an internal control and use normalization to account for variability across samples. Both transient transfection and stable cell lines are used, with adaptations for high-throughput screening when needed.

Applications of reporter assays include characterizing promoter and enhancer activity, assessing transcription factor or signaling pathway activity, and performing drug or compound screening. In vivo versions exist, using transgenic animals or viral delivery. Limitations include differences between plasmid-based context and chromosomal regulation, variable transfection efficiency, reporter maturation times, and potential artifacts from overexpression or altered cellular states.
