Receptor-mediated refers to cellular processes that require the specific interaction between a ligand and its receptor to initiate a response. This term encompasses mechanisms such as receptor-mediated endocytosis and receptor-driven signaling, where binding events confer selectivity and regulate downstream outcomes. It stands in contrast to non-specific uptake pathways that do not rely on defined receptor-ligand interactions.

In receptor-mediated endocytosis, a ligand binds to its cell-surface receptor, prompting the receptor to cluster in specialized membrane domains such as clathrin-coated pits. The plasma membrane invaginates to form vesicles that internalize the ligand-receptor complex. Inside the cell, vesicles fuse with early endosomes, where a change in pH can cause ligand release. Receptors are often recycled to the plasma membrane, while some ligand-receptor complexes are directed to lysosomes for degradation. This mechanism enables cells to acquire nutrients and regulate signaling. Examples include the LDL receptor pathway for cholesterol uptake, the transferrin receptor for iron transport, and the insulin receptor for glucose metabolism.

Receptor-mediated signaling involves ligands binding to receptors that either possess intrinsic enzymatic activity or associate with other signaling proteins. This triggers cascades such as the receptor tyrosine kinase pathways (e.g., EGFR), G protein-coupled receptor pathways, and cytokine receptor–activated routes like JAK-STAT. The resulting signals influence gene expression, metabolism, growth, differentiation, and survival.

Applications include drug development and targeted therapies. Abnormal receptor function can contribute to diseases, while interventions—such as monoclonal antibodies, small-molecule inhibitors, or ligand-drug conjugates—aim to modulate receptor-mediated processes for therapeutic benefit.
