pMB1-based refers to plasmids that use the pMB1 origin of replication. The pMB1 origin is a derivative of the ColE1-type replication system and is widely used in bacterial cloning vectors because it tends to produce a relatively high copy number in Escherichia coli, enabling abundant plasmid DNA and often high-level expression of cloned genes.

Vectors described as pMB1-based are common backbones for molecular cloning. They typically feature a selectable marker such as an ampicillin resistance gene, a multiple cloning site for inserting foreign DNA, and often a screenable marker such as lacZ alpha for blue/white screening. The exact copy number of a pMB1-based plasmid depends on the host strain, growth conditions, and specific vector design; in practice, these plasmids range from moderate to very high copy numbers.

The pMB1 origin is part of the larger ColE1 family and is regulated through RNA-based mechanisms that balance replication and stability, a characteristic shared with many ColE1-type origins. Because many plasmids use the ColE1/pMB1 origin, compatibility must be considered when co-transforming with other plasmids to avoid replication interference.

In use, pMB1-based vectors are favored when high yields of plasmid DNA are required or when high-level expression of cloned inserts is desired. However, high copy can impose a metabolic burden on the host and affect plasmid stability in some strains.
