OppervlakteELISA, also known as Surface ELISA (Enzyme-Linked Immunosorbent Assay), is a variant of the ELISA technique specifically designed to detect and quantify antigens or antibodies bound to the surface of cells, particles, or solid substrates. Unlike traditional ELISA, which relies on immobilized antigens or antibodies in a microplate, Surface ELISA focuses on interactions occurring at the cell surface or on the surface of other biological or synthetic particles.

In this method, cells or particles are first immobilized on a solid surface, such as a microplate well or a membrane. The target antigen or antibody is then allowed to bind to its specific receptor or ligand on the cell surface. Following this, a detection antibody conjugated with an enzyme is introduced. After washing to remove unbound components, a substrate is added that reacts with the enzyme to produce a measurable signal, typically colorimetric, fluorescent, or chemiluminescent.

Surface ELISA is particularly useful in studying cell surface proteins, receptor-ligand interactions, and immune responses at the cellular level. It is widely applied in immunology, virology, and cell biology research. The technique can also be adapted for high-throughput screening, allowing for the simultaneous analysis of multiple samples. However, it requires careful optimization to minimize non-specific binding and ensure accurate detection of surface-bound molecules.

One advantage of Surface ELISA is its ability to provide quantitative data on cell surface markers without the need for cell lysis, preserving the integrity of the sample. This makes it a valuable tool for analyzing live cells or particles directly. Variations of the method include competitive and sandwich assays, depending on the experimental requirements. Despite its utility, Surface ELISA may face challenges such as background noise and the need for precise surface immobilization techniques.
