Nonproteoglycan is a term used to refer to molecules that are not proteoglycans. In the context of the extracellular matrix (ECM) and cell surfaces, nonproteoglycans encompass a wide range of proteins and protein assemblies that lack the characteristic glycosaminoglycan (GAG) chains attached to a core protein. By contrast, proteoglycans are glycoproteins heavily substituted with GAG side chains, which confer high negative charge, hydration, and space-filling properties.

Major examples of nonproteoglycans include structural ECM proteins such as collagens (providing tensile strength), elastin (imparting elasticity), and the adhesive glycoproteins fibronectin and laminin, as well as matricellular proteins such as tenascin and thrombospondin. Other nonproteoglycans include various cell-surface receptors (e.g., integrins), enzymes (such as matrix metalloproteinases), and other secreted glycoproteins that regulate adhesion, signaling, and matrix remodeling. Many nonproteoglycans interact with proteoglycans, forming composite networks that influence tissue architecture and signaling dynamics.

Because the ECM is a complex mixture, the distinction between proteoglycans and nonproteoglycans is not always strict at the functional level. Nonproteoglycans contribute to mechanical strength, organization, and cell communication, while proteoglycans modulate hydration, charge distribution, and growth factor presentation. Dysfunctions in nonproteoglycan components can contribute to fibrosis, osteoarthritis, cancer progression, and wound healing defects.
