Mikrotubulusnucleációs refers to the process by which microtubules are initiated and assembled. Microtubules are essential components of the cytoskeleton, playing roles in cell shape, intracellular transport, and cell division. Their formation is a highly regulated process, and it begins with the formation of a small, stable seed structure. This seed is typically composed of gamma-tubulin, which is found in a ring complex. These gamma-tubulin ring complexes act as nucleation sites, recruiting alpha- and beta-tubulin heterodimers. These heterodimers then assemble onto the growing end of the microtubule, a process known as polymerization.

The primary site of microtubule nucleation in eukaryotic cells is the centrosome. The centrosome contains two centrioles and a pericentriolar matrix, which is rich in gamma-tubulin ring complexes. From the centrosome, microtubules radiate outwards to form the mitotic spindle during cell division and the cytoplasmic microtubule network in interphase cells. However, microtubule nucleation is not solely confined to the centrosome. Non-centrosomal nucleation can occur at various locations within the cell, often associated with specific cellular structures or signaling pathways. This allows for the formation of microtubules in particular regions where they are needed. The rate of microtubule polymerization and depolymerization is influenced by various factors, including the concentration of free tubulin heterodimers and the presence of microtubule-associated proteins (MAPs). This dynamic regulation is crucial for the diverse functions of microtubules.
