Melanosomer is not a standard term in modern cell biology. The organelle responsible for pigment synthesis and storage in pigment cells is usually called the melanosome. Melanosomes are found in melanocytes of the skin and hair follicles, as well as in retinal pigment epithelium, and they play a central role in coloration and protection from ultraviolet radiation.

Melanosomes are membrane-bound organelles that originate in the endosomal-lysosomal system. They go through a maturation process classified into stages I through IV, during which pigment-synthesizing enzymes such as tyrosinase, TYRP1, and DCT are acquired and melanin is deposited within a specialized matrix formed by PMEL. The internal environment becomes progressively more conducive to melanin synthesis as the pH and enzymatic activity change.

Biogenesis and trafficking: Melanosomes form in early endosomes of melanocytes and mature as they are transported to the cell periphery along dendritic extensions. Transport depends on motor and adaptor proteins including Rab27a, melanophilin (Slac2-a), and myosin Va, which guide melanosomes toward keratinocytes or other recipient cells in pigmented tissues.

Clinical relevance and variation: Defects in melanosome formation, maturation, or transport underlie several pigmentary disorders. Examples include Hermansky-Pudlak syndrome and Chediak-Higashi syndrome, both caused by mutations in components of the lysosome-related organelle system, leading to hypopigmentation and other symptoms. In humans, natural variation in melanosome biology contributes to the wide range of skin, hair, and eye colors observed across populations. In summary, while melanosome is the standard term, melanosomer may appear in older literature or as a misspelling.
