Megakaryocytes are large bone marrow cells. They are the largest cells found in the bone marrow. Their primary function is to produce platelets, which are essential for blood clotting. Megakaryocytes develop from hematopoietic stem cells through a process of proliferation and differentiation. This process involves several stages, including the formation of pro-megakaryocytes and then megakaryocytes.

The cytoplasm of a mature megakaryocyte is abundant and contains granules that will eventually become part of the platelets. These granules store various molecules important for hemostasis and wound healing. Once mature, the megakaryocyte extends long cytoplasmic extensions, called proplatelets, into the bloodstream or sinusoids of the bone marrow. These proplatelets fragment into individual platelets.

Megakaryopoiesis, the process of megakaryocyte development and platelet production, is tightly regulated by various growth factors and cytokines, with thrombopoietin being the most significant regulator. Disorders affecting megakaryocytes can lead to abnormalities in platelet count, such as thrombocytopenia (low platelet count) or thrombocytosis (high platelet count), impacting an individual's ability to stop bleeding. Megakaryocytes are a critical component of the hematopoietic system, underpinning the vital function of hemostasis.
