In pharmacology, mechanism-based inhibition, also called suicide inhibition, refers to inhibitors that are processed by an enzyme into reactive species that covalently modify and permanently inactivate the enzyme. Classic examples include allopurinol, which is converted by xanthine oxidase into a reactive form that inactivates the enzyme, and aspirin, which irreversibly acetylates the active site of cyclooxygenase. Such inhibition typically requires the target's catalytic activity, conferring high selectivity but raising safety considerations due to potential off-target metabolism.

In drug discovery and chemical biology, mechanism-based design or mechanism-based therapy uses knowledge of reaction mechanisms, transition states, or catalytic steps to craft inhibitors, activators, or probes. Strategies include transition-state mimics and covalent inhibitors that form durable, mechanism-dependent interactions. Mechanism-based approaches can help distinguish closely related enzymes but may demand detailed structural and kinetic information.
