Lactose-utilizing refers to the capacity of an organism or cell to metabolize lactose, a disaccharide composed of glucose and galactose, as a carbon and energy source. This ability varies across organisms: many bacteria and some yeasts can actively import and break down lactose, while in humans lactose is typically hydrolyzed by intestinal lactase to permit absorption rather than directly metabolized.

In bacteria, lactose utilization is commonly governed by a system known as the lac operon, which in its classic form includes lacZ (beta-galactosidase), lacY (lactose permease), and lacA (a transacetylase). Lactose or the inducer allolactose binds to the LacI repressor, enabling transcription of the operon. Imported lactose is cleaved by beta-galactosidase into glucose and galactose; glucose enters glycolysis, and galactose is processed via the Leloir pathway to feed central metabolism. Regulation is influenced by the availability of other carbon sources; in the presence of preferred sugars like glucose, lactose utilization can be repressed through catabolite repression.

Beyond Escherichia coli, other microbes such as Lactobacillus and Bifidobacterium species utilize lactose through diverse permeases and beta-galactosidases, contributing to dairy fermentation and gut ecology. In industry, organisms are often engineered to enhance lactose utilization for dairy processing, lactose-free product production, or bio-based conversions.

In humans, lactose utilization relates to lactase persistence or deficiency. Lactase activity in adulthood affects digestion of dairy products; in lactose malabsorption, undigested lactose can be metabolized by gut microbes, sometimes causing gastrointestinal symptoms. Understanding lactose-utilizing traits informs microbiology, food science, and metabolic engineering.
