Interstitiviruses are a group of small, non-enveloped viruses that infect a wide range of hosts, including humans, animals, and plants. They are characterized by their ability to replicate within the cytoplasm of infected cells, rather than in the nucleus. This group includes several well-known viruses, such as the hepatitis delta virus (HDV), the hepatitis E virus (HEV), and the human parvovirus B19 (B19V).

The term "interstitivirus" is derived from the Latin word "interstitium," meaning "between spaces," reflecting the viruses' preference for replicating in the intercellular spaces of tissues. This characteristic is thought to contribute to their ability to evade the host's immune response and persist in infected individuals.

Interstitiviruses are typically small, with genomes ranging from 1.7 to 3.5 kilobases in length. They encode a single polyprotein that is processed into functional proteins by viral and host proteases. The replication strategy of interstitiviruses involves the synthesis of a negative-sense RNA genome, which is then used as a template for the production of positive-sense RNA molecules.

Interstitiviruses are known to cause a variety of diseases in their hosts. For example, HDV is a satellite virus that requires the presence of hepatitis B virus (HBV) for replication and can cause severe liver disease in infected individuals. HEV is a major cause of acute hepatitis in humans, particularly in developing countries, and can lead to fulminant hepatitis and death. B19V is a common cause of erythema infectiosum, a mild rash, and can also cause more severe diseases, such as aplastic anemia and arthritis.

The study of interstitiviruses has been facilitated by the development of molecular techniques, such as reverse transcription polymerase chain reaction (RT-PCR), which allow for the detection and characterization of these viruses in clinical samples. However, the lack of a suitable animal model for interstitivirus infection has hindered the development of effective antiviral therapies and vaccines. Ongoing research aims to address these challenges and improve our understanding of these important pathogens.
