Immunobeads are small particles, typically microspheres, coated with antibodies or other ligands that selectively bind target biomolecules. They are used to capture, enrich, or remove specific antigens or cells from complex samples. Beads are available in magnetic and non-magnetic forms; magnetic beads can be rapidly separated from solution with an external magnet, enabling quick washing and processing, while non-magnetic beads require centrifugation or filtration.

Common bead materials include polystyrene, silica, and iron oxide cores. Surfaces are functionalized to enable covalent attachment of antibodies or ligands, using chemistries such as NHS-ester, carbodiimide (EDC), epoxy, carboxyl, or streptavidin-biotin linkages. Antibodies may be monoclonal or polyclonal; ligands can also be aptamers, Protein A/G, or other capture molecules. Binding efficiency can be influenced by antibody orientation and surface density.

Applications span immunoprecipitation and pull-down assays, immunomagnetic separation of cells, and purification or depletion of proteins, nucleic acids, viruses, or extracellular vesicles from biological samples. Beads are also used as capture reagents in multiplex bead-based assays and diagnostic tests. A typical workflow involves incubating sample with immunobeads, separating bound complexes, washing away unbound material, and eluting or directly analyzing the captured targets.

Advantages include high specificity, compatibility with complex matrices, and rapid processing. Limitations include nonspecific binding, the need for careful optimization of binding conditions, potential antibody leakage, and cost. Batch-to-batch variability and the requirement for appropriate controls should be considered in experimental design.
