Glucuronides are a class of molecules formed by the conjugation of glucuronic acid with other molecules. This process, known as glucuronidation, is a crucial metabolic pathway in many organisms, particularly in the liver, and serves to detoxify and facilitate the excretion of various endogenous and exogenous compounds. Glucuronic acid is a derivative of glucose, and its conjugation to other substances increases their water solubility, making them easier for the body to eliminate, primarily through urine and bile.

The formation of glucuronides is catalyzed by a family of enzymes called UDP-glucuronosyltransferases (UGTs). These enzymes attach glucuronic acid, in the form of its activated precursor UDP-glucuronic acid, to a wide range of substrates. These substrates include bilirubin, a breakdown product of heme; steroid hormones; thyroid hormones; and numerous drugs and environmental toxins. For instance, many pharmaceutical medications undergo glucuronidation as a primary metabolic route, influencing their duration of action and elimination from the body.

Glucuronidation plays a significant role in detoxification by converting potentially harmful lipophilic (fat-soluble) compounds into more hydrophilic (water-soluble) forms. This transformation prevents their accumulation in tissues and facilitates their removal. Disorders affecting glucuronidation pathways, such as certain genetic conditions, can lead to the buildup of toxic substances in the body, resulting in various health issues. The study of glucuronides is therefore important in pharmacology, toxicology, and medicine.
