Flourescentielabeling is a labeling technique in which fluorescent probes are attached to biological molecules to enable visualization with light-based detection methods. The approach enables researchers to locate, track, and quantify biological structures and processes in cells, tissues, and in vitro assays. Labels are chosen for their brightness, stability, and spectral properties to suit specific imaging needs.

Common fluorescent labels include small molecule dyes such as fluorescein, rhodamine, and cyanine dyes; fluorescent proteins like GFP and mCherry; and inorganic or semi-synthetic probes such as quantum dots and lanthanide chelates. Labeling can be achieved through covalent attachment to amino, carboxyl, or sulfhydryl groups, often using NHS esters, maleimide chemistries, or click chemistry. Enzymatic or genetic methods, such as SNAP-tag, HaloTag, or expression of fluorescent protein fusions, enable site-specific labeling in living cells.

Labeling strategies fall into several categories: covalent chemical labeling of biomolecules, enzymatic tagging that generates a fluorescent handle, and genetic encoding of fluorescent reporters. Each method offers different control over labeling stoichiometry, specificity, and compatibility with live-cell imaging.

Applications are widespread and include fluorescence microscopy, flow cytometry, immunofluorescence, and in situ hybridization. Fluorescent labeling supports dynamic studies, multiplexed detection, and quantitative measurements, but it is limited by photobleaching, spectral overlap, and potential alteration of native function upon labeling.

Safety and experimental design considerations are important, including label toxicity, fixation effects, and appropriate controls to distinguish signal from background.
